efficient one-pot enzymatic synthesis of trehalose 6-phosphate. The one-pot reaction of trehalose 6-phosphate phosphorylase and maltose phosphorylase enables production of 65 mM trehalose 6-phosphate from 100 mM maltose, 100 mM Glc6P, and 20 mM inorganic phosphate. Addition of beta-phosphoglucomutase to this reaction produces glucose 6-phosphate from beta-glucose 1-phosphate and thus reduces requirement of glucose 6-phosphate as a starting material. Within the range of 20-469 mM inorganic phosphate tested, the 54 mM concentration yielded the highest amount of trehalose 6-phosphate (33 mM). Addition of yeast increases the yield because of its glucose consumption. From 100 mmol maltose and 60 mmol inorganic phosphate, the production of 37.5 mmol trehalose 6-phosphate in a one-pot reaction (100 ml) is achieved, and 9.4 g trehalose 6-phosphate dipotassium salt is obtained
